Lady Anne Spencer was the eldest daughter of Robert, 2nd Earl of Sunderland. In 1687 she married James, Earl of Arran, the eldest son of William, 3rd Duke of Hamilton and Anne, Duchess in her own right.
He did not inherit the titles on his father's death in 1694 but only when his mother Duchess Anne resigned them in his favour in 1698.
Lady Anne bore her husband two daughters, both of whom died young. She herself died at Kinneil, of puerperal fever, in her 24th year and was buried at Hamilton.
